Antispasmodic effect of 4'-methylepigallocatechin on guinea pig ileum.

AbstractThe antispasmodic effect of 4′-methylepigallocatechin (MEC), which was isolated from Maytenus rigida Mart (Celestraceae), was investigated in vitro in guinea pig intestinal segments. In the isolated ileum, MEC (1 nM–100μM) did not modify the ileal spontaneous tonus or the electrically elicited contractions. MEC (8μM) significantly (p<0.01) reduced the submaximal contractions induced by histamine (2μM), carbachol (100μM) and BaCl2 (0.03M). An additive relaxing action (p<0.001) was observed by co-incubation of verapamil (10 nM) and MEC (8μM). Although MEC (1nM–100μM) did not modify the contractions elicited by 60mM KCl, it significantly reduced the CaCl2 contractile response without c…

Die Substanzen zeigten am isolierten Rattendarm eine kompetitiv-nichtkompetitive anticholinerge Wirkung, am isolierten Meerschweinchendarm eine kompetitiv-nichtkompetitive Antihistaminwirkung. An isolierten Meerschweinchenvorhofen trat nach Gabe der tertiaren 2-Skatyl-alkylpyridine eine durch Atropin aufzuhebende negativ inotrope Wirkung auf, die quartaren Substanzen waren dagegen positiv inotrop wirksam. Cannabinoid CB1 receptor activation modulates spontaneous contractile activity in mouse ileal longitudinal muscle.

The purpose of the present study was to examine whether cannabinoid receptor agonists influence spontaneous contractile activity of longitudinal muscle in mouse ileum in vitro. Isolated segments of mouse ileum displayed spontaneous contractions with an amplitude and frequency of about 300 mg and 30 cpm, respectively. The endocannabinoid anandamide (1-100 microM), the selective cannabinoid CB(1) receptor agonist, ACEA (0.1 microM-10 microM), but not the selective cannabinoid CB(2) receptor agonist, JWH 133 (0.1 microM-10 microM), reduced in a concentration-dependent manner the spontaneous mechanical activity. 5-Oxo-1,4-dihydroindenopyridines: Calcium modulators with partial calcium agonistic activity

The title compounds 4a-g were prepared by Knoevenagel reactions of 1,3-indandione (1) with the aromatic aldehydes 2a-g followed by cyclizing Michael addition of the products thus obtained with methyl β-aminocrotonate (3). The structures of products 4 were characterized by spectal data. Positive inotropic activities were observed on electrically stimulated, left atria of giunea pigs and these effects could be attributed to calcium agonism. On the other hand, barium chloride-induced contractions of guinea pig ileum were inhibited in a dose-dependent manner.
